Answer: Whether an officer is on duty or not on the day of a trial is irrelevant when it comes to whether they can testify. If the officer has been called by the court to appear or testify, he can testify no matter if he is on duty or not. He could be required if the court has issued a subpoena for him to testify or willingly come to court to testify.
